In this paper we propose a model that determines the activation of synapse while the nerve impulse is spreading throughout. In particular we investigate presynaptic membrane that holds vesicles with the mediator. It injects into the synaptic cleft when the nerve impulse arrives. It is taken into account that vesicles are grouped in several pools. The mediator injection in the cleft depends on which pool the vesicle holds. In the model we also account pool-to-pool transitions of vesicles.
